年少即至蘇州，學習崑曲，以身段柔軟，動作滑稽聞名。後入燕京，改唱京劇，也受到喜愛，時稱「蘇丑第一」，武戲也非常傑出，名震京師。
楊鳴玉過世後一年，正值甲午戰爭失利，李鴻章簽訂馬關條約，從此福建臺灣省割讓予日本，有人以輓聯一副輓之：“楊三已死無蘇丑，李二先生是漢奸”。這個無情對廣傳天下，上聯輓楊鳴玉，下聯卻是諷刺人稱「李二」的李鴻章。也有說法「楊三」是名丑劉羊三的訛寫。
